The current study attempts to examine the potential mechanism of magnolol and its protective effect in reducing the progression of experimental OA in vivo and in vitro. In vitro, nitric oxide (no), prostaglandin E2 (PGE2), cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), tumor necrosis factor α (TNF- α) And interleukin-1 β (IL-1 β) Induced IL-6 was significantly inhibited by magnolol in a concentration dependent manner. In addition, magnolol may also down regulate the expression of metalloproteinase 13 (MMP13) and platelet reactive protein motif 5 (adamts5). Mechanically, magnolol inhibits PI3K / Akt / NF- κ Activation of B pathway. In addition, our molecular docking study also revealed the strong binding ability between magnolol and PI3K. In addition, the protective effect of magnolol on the development of OA was detected in mouse model.
